Melleril
Contents:
Thioridazine hydrochloride
Dosage:
Anxiolytic & antidepressant
Adult 10-75 mg daily.
Children ( In addition :
difficulties of concentration, behavioural disorders, hyperactivity,
aggressiveness, sleep disturbances ) 0.5-2 mg/kg body weight daily in
divided doses.
Neuroleptic
Hospitalised psychotics
100-600 mg.
Psychotic outpatients
50-300 mg.
Alcoholic patients during
withdrawal 100-200 mg.
Severe neuroses
25-150 mg. Max : 800 mg daily.
Children ( Childhood
schizophrenia, severe behavioural disorders in mental retardation,
psychopathy ) 1-4 mg/kg body weight daily in divided doses.
Contraindications:
Comatose states or severe central
nervous system (CNS) depression; history of blood dyscrasias; severe
cardiovascular (CV) disease.
Special Precautions:
Narrow-angle glaucoma, prostatic
hypertrophy, CV disease, impaired liver function; when driving a vehicle or
operate machinery; regular blood counts during the 1st few month &
immediately when symptoms suggestive of blood dyscrasia occur;
anticonvulsant medication may need dosage adjustment; pregnancy & lactation.
Adverse Reactions :
Sedation, vertigo, orthostatic
hypotension, nasal stuffiness, dry mouth, disturbances of visual
accomodation, urinary retention or incontinence, galactorrhea, menstrual
irregularities, disturbances of ejaculation, skin rashes, photosensitivity,
nausea, constipation, weight change, hyperthermia, jaundice. At high dose
extraphyramidal symptom; very rarely neuroleptic malignant syndrome, tardive
dyskinesia after prolonged treatment. ECG changes, rarely arrhythmia, sudden
unexplained death. Pigmentary retinopathy ( with very high doses & long-term
treatment).
Drug interactions :
Alcohol, CNS depressants,
antihistomine, anticholinergics quinidine; levodopa, anticonvulsants,
adrenergic vasoconstrictors; MAO inhibitors, lithium, β-blockers,
antacids, anti-diarrheal drugs.
