Sinemet/Sinemet 25/100
Contents:
Per Sinemet tab Carbidopa 25 mg,
levodopa 250 mg.
Per Sinemet 25/100tab Carbidopa
25 mg, levodopa 100 mg.
Indications:
Treatment of symptoms of
parkinsonism.
Dosage:
Sinemet 25/100 1 tab three
times daily. Max: 8 tab.
Sinemet 25/250 ½ tab once-bd.
Max 8 tab.
Transfer from levodopa
Stop levodopa therapy 12 hourly before Sinemet is started ( 24 hours for SR
preparation ).
Patients taking
≤ 1500 mg levodopa Initial: 1 tab Sinemet 25/100 three-four times daily, >
1500 mg levodopa Inital: 1 tab Sinemet three-four times daily.
Contraindications:
Narrow-angle glaucoma.
Undiagnosed skin lesions or history of melanoma. Hypersensitivity to any
component of this product.
Special Precautions:
Cardiovascular (CV) diseases
including history of myocardial infarction (MI) or arrhythmias or pulmonary
disease, bronchial asthma, renal, hepatic o endocrine disease, history of
psychoses or convulsions, or wide-angle glaucoma. Absorption of I-dopa may
be impaired in some patients on a high protein diet. Not recommended for
children
≤ 18 years.
Adverse Reactions:
Dyskinesia including choreiform,
dystonic & other involuntary movements. Mental changes including paranoid
ideation & psychotic episodes; depression with or without development of
suicidal tendencies; dementia; & gastrointestinal (GI) disturbances.
Drug interactions:
Discontinue MAOIs 2 weeks prior
to therapy with Sinemet ( except low doses of MAO-B inhibitors ), tricyclic
antidepressants, antihypertensive drugs.
